After spraying it three times, its death animation will play, after which the shine will spawn. You should position yourself in a way that will allow you to collect the shine quickly once it spawns. You can do this by lining up with your back toward the plant, then when steam starts appearing, you can hold R down all the way then hit A to do a backflip.
You need the spray nozzle equipped to do this. If you lined it up correctly, Mario will land in the right place as the cutscene starts playing. After the shine spawn animation finishes, hit A and then L immediately after to do a short ground pound and collect the shine.
